The poor oral bioavailability contributed by the insolubility and rapid biotransformation has been established as the major limitation for the ‘functional’ or ‘nutraceutical’ applications of curcumin. Though a number of bioavailable formulations with enhanced bioavailability of ‘curcumin metabolites’ have been reported and available commercially as curcumin supplement, oral delivery forms capable of providing curcuminoids in its native bioactive forms as ‘free (unconjugated)’ curcuminoids remains as a challenge.
Recent understanding on the relative bioactivity and permeability of ‘free’ curcuminoids have demonstrated that curcuminglucuronides (major conjugated metabolites) possess no anticancer, anti-inflammatory and antioxidant activities (Bioorg. Med. Chem. 2014, 22, 435; Food Chem, 2014, 151, 126; Antioxidants, 2015, ISSN-2937) and low membrane permeability(J Pharmacol. Exptl Therapeutics, 2008, 326, 196). Thus, the key factor responsible for the efficacy of a curcumin formulation is turned out to be the “free curcuminoids availability” in plasma and its cellular distribution. The mere number of folds of bioavailability as determined by the area under curve of curcumin metabolites in plasma as reported so far became questionable.
CurQfen is a bioavailable curcumin, capable of providing ‘free’ curcuminoids into plasma and further into organ tissues with improved BLOOD-BRAIN-BARRIER permeability ( Journal of Functional Foods, 2015, 14, 215 ).
